| خلاصه مقاله | Background and Aims Drug-excipient compatibility study is essential for excipient selection at the pre-formulation stage (1). Kojic acid is one of the skin-lightening agents due to its tyrosinase inhibitory activity (2). Propylene glycol uses as emulsifiers, cleansing agents, humectants, and skin conditioners (3). The recent research investigated the compatibility between kojic acid and propylene glycol utilizing both thermal and non-thermal techniques. - Materials and Methods The blend of kojic acid and propylene glycol was prepared by combining them in equal proportions (1:1) with a vortex mixer. Then, 20% w/v water was added and thoroughly mixed. These samples were then kept in an oven at 40 °C for two months. Compatibility of the mixture was assessed through DSC and FT-IR techniques (4). Additionally, a validated HPLC-UV analytical method was utilized to measure the remaining kojic acid post-stress (5). The ethical code was TBZMED.REC.1394.496. - Results The combinations of kojic acid and propylene glycol in the mixtures exhibited a noticeable color change. The DSC pattern of the blend showed a new endothermic peak at the temperature of 119 °C, which suggested a possible alteration in the formulation. Despite this, the FT-IR analysis indicated that kojic acid remained unaffected by propylene glycol during stress test, due to the limited sensitivity of the FT-IR method. Additionally, the HPLC results indicated that approximately 56.53 ± 1.24% of kojic acid remained in the samples after the stress test. Both the HPLC and DSC results consistently supported each other in their findings. - Conclusion The DSC and HPLC analyses confirmed that kojic acid and propylene glycol are not compatible. It is recommended to avoid using propylene glycol as a skin conditioner in cosmeceuticals that contain kojic acid. |
