| خلاصه مقاله | Introduction
The significance of immunotherapy in the systemic therapy of hepatocellular carcinoma (HCC) is escalating as our comprehension of the liver suppressive mechanism and tumor microenvironment advances (1). Although Immune checkpoint inhibitors have exhibited substantial clinical results in HCC patients, extensive in-depth studies into the molecular pathways linked with their therapeutic efficacy are still necessary to discover more effective strategies (2). The purpose of this study was to investigate the effect of small interfering RNA (siRNA)-mediated silencing of programmed death-ligand 1 (PD-L1) on HCC cells apoptosis and the implicated genes.
Materials and Methods
Following PD-L1 siRNA transfection of HepG2 cells and RT-PCR analysis for transfection efficiency, both transfected and untransfected cells underwent flow cytometry to assess apoptosis rate. The expression levels of the apoptotic genes BAX and CAS9, as well as the anti-apoptotic gene BCL2, were then measured using RT-PCR to investigate the underlying mechanism.
Results
RT-PCR analysis revealed a significant decrease in PD-L1 mRNA expression levels in HepG2 cells after PD-L1 siRNA transfection. The flow cytometry analysis of apoptosis then demonstrated that PD-L1 siRNA could substantially increase the number of apoptotic HepG2 cells. Ultimately, the RT-PCR analysis of PD-L1 siRNA-transfected HepG2 cells found that the apoptotic genes BAX and CAS9 were significantly upregulated, while the anti-apoptotic gene BCL2 was significantly downregulated.
Conclusions
We found that siRNA-mediated PD-L1 knockdown enhances apoptosis in HCC by upregulating apoptotic genes while downregulating antiapoptotic genes. |
