| خلاصه مقاله | Introduction: Hepatocellular carcinoma (HCC) is a highly frequent primary liver tumor and the world's third largest cause of cancer death. Autophagy is a conserved biological function that is essential for the maintenance of homeostasis and the reduction of metabolic stress. Autophagy has recently been the focus of much attention due to its dual role in cancer progression and inhibition. This metabolic process is associated with poor survival in hepatocellular carcinoma and is one of the major causes of drug resistance. Inhibition of some autophagy genes can be effective in suppressing advanced HCC. On the other hand, induction of autophagy can increase the sensitivity of HCC to chemotherapy drugs. Of note, many conventional clinical treatments face challenges such as drug toxicity and off-site cancer targets. Nanoparticles have largely overcome these obstacles due to their ability to target delivery and control the release of autophagy regulators. This review focuses on the use of nanoparticles for combination therapy by targeting autophagy in hepatocellular carcinoma treatment. Methods: Original articles from 2015 to the present targeting of autophagy process in hepatocellular carcinoma were reviewed from Google Scholar, Web of Science, and PubMed databases. Results: Despite the obstacles to using autophagy modulatory agents in combination therapies, nanotechnology-enabled autophagy modulatory combination therapy may be an effective strategy in the treatment of hepatocellular carcinoma due to enhanced drug blood circulation, little side effect, and enhanced stability, and solubility of drugs and regulators. For instance, lipid coated calcium carbonate and self-assembled poly (ethylenimine) Glycyrrhetinic acid nanoparticles were employed for combination therapy using autophagy modulators miR375 and shRNA silencing Akt1 and chemotherapy agents against hepatocellular carcinoma. Conclusion: Utilizing combination chemotherapy and autophagy inhibitors/inducers loaded on nanocarriers helps to increase drug sensitivity and the efficacy of the treatments employed targeting hepatocellular carcinoma. |
