| خلاصه مقاله | Feeding infants with safe milk from the beginning of their life is an important issue. Drugs which are used by mothers can affect the composition of milk in a way that is not only unsuitable, but also toxic for infants. The aim of this study is to find out the relationship between milk/plasma ratio of drugs and their chemical structures and to develop mechanistic models for predicting the penetration of drug into human milk. Milk to plasma (M/P) concentration ratio of drugs was used to categorize drugs into high risk and low risk. For each drug, different structural parameters, including logD7.4, polar surface area (PSA) and different functional groups count, were calculated and applied to develop different interpretable and mechanistic models for milk/plasma ratio prediction of drugs during breastfeeding period. The results of this study show that the number of hydrogen bond acceptors, number of acidic oxygen, polar surface area and total surface area are among the most vital structural parameters for predicting. Developed models of this study can be useful for predicting milk/plasma ratio for new drug candidates during breastfeeding period with an acceptable error. |
