خلاصه مقالهBackground and Aim: Deviation in immune response and dysregulation in cytokine profiles have been reported in subjects with autism spectrum disorder (ASD). The relative quantity of cytokines was determined in the peripheral blood mononuclear cells (PBMC) of ASD children after primary vaccination against Hepatitis B virus (HBV) and after vaccine booster challenge. Methods: PBMC samples were prepared before and after booster injections from 16 autistic children in comparison with 4 healthy subjects. Total RNA were extracted and the cDNA were synthesized. The relative quantities of transcripts for cytokines were assessed. Results: The mean age of participants was 7.6 ± 2.5. Both groups responded similarly to the booster vaccine. Before booster injection, levels of IFN-γ were somehow similar between both groups. The relative mean values for IL-4, IL-10 and IL-12 in ASD were higher; however, TGF-β relative mean levels were significantly lower than that of controls. One month after injection, in ASD, the relative mean of IL-4 up regulated slightly. IL-10 was decreased to control group values. TGF-β value was increased in both groups with no major difference and the levels of expression for IL-12 and IFN-γ were significantly increased in ASD patients. IL-17 mRNA was not detected in PBMCs from ASD and control groups. Before booster, Th-2 type cytokines were significantly higher in ASD patients whereas, after booster injection the Th-1type significantly increased in comparison with control subjects. Conclusion: These results indicated a potential role of immune dysfunction in ASD which would assist discovering of novel possible therapeutics for this condition.
