خلاصه مقالهObjective To maintain fertility before gonadotoxic therapy, ovarian tissue can be frozen and transplanted again after therapy, but not in patients with a medium / high risk of ovarian metastasis. The artificial ovary is a promising alternative, because the ovarian follicles are extracted from the tissue and thus separated from malignant cells. An essential factor in the development of the artificial ovary is a protocol that extracts as many ovarian follicles as possible intact and capable of survival. Material and methods A comparative analysis of purely mechanical and combined mechanical-enzymatic extraction methods of porkine ovarian follicles was carried out, whereby the concentrations of the two enzymes used (collagenase, liberase) and the enzyme treatment protocol used were varied. To evaluate the viability, intact follicles were cultivated for 10 days with a subsequent live / dead assay for vitality analysis [ Figure 1 ]. [1] Results The number of intact follicles was higher when using collagenase (421) than with Liberase (393). The viability of the follicles was higher for the Liberase extraction: after 10 days of cultivation, 92.45% of 138 follicles with Liberase extraction were vital (> 90% living granulosa cells) and 82.14% of 127 follicles with collagenase extraction. Intact follicles could not be extracted with purely mechanical extractions. Summary A combination of mechanical and enzymatic (0.5 mg / ml Liberase concentration) extraction proved to be the most productive extraction method. The protocol determined can serve as a basis for in-vitro and in-vivo testing of the artificial ovary
