| Breast cancer (BC) is the second most common cause of cancer-related deaths in women worldwide. Twist1 and Twist2 belong to a type of transcription factors involved in metastasis that are affected by promoter methylation in CpG islands. Hypermethylation of these genes induces epithelial-mesenchymal transition (EMT) through the negative regulation of E-cadherin (E-cad) and promotes cell invasion. The present study was designed to assess the methylation status of Twist1 and Twist2 in patients with BC. After bioinformatics analysis of the mentioned genes and collecting the breast tumors and their corresponding margin samples from 83 patients, the methylation status of the Twist1 and Twist2 genes was evaluated using methylation-specific PCR and gel electrophoresis. Finally, the results were analyzed according to the McNemar and Chi-square tests (P ≤ 0.05). The results showed considerable methylation of both Twist1 and Twist2 genes in tumors, and to a lesser extent in margins. Furthermore, significant correlations have been found between methylation status and clinicopathologic characteristics of the patients (P < 0.001). In addition, we found that the promoter regions of Twist1 and Twist2 were frequently methylated in BC that showed significant correlations with the clinicopathologic characteristics of the patient. Based on these findings, the methylation modification of Twist1 and Twist2 could be considered potent prognostic biomarkers for patients with BC. |
