| Obesity is associated with numerous metabolic and inflammatory disorders. This study aimed to evaluate the effects on
hepatic oxidative stress and inflammation in a rat model of obesity induced with a high-fat diet, and to investigate the
effects of vitamin D administration on markers of oxidative stress and metabolic damages of liver. Twenty rats received a
normal diet and 20 a high fat diet for 16 weeks. At the end of this period, a subgroup of 10 rats within each group received
500 IU/kg vitamin D for 5 weeks; the other 10 served as control. The concentrations of tumor necrosis factor, glutathione
peroxidase, superoxide dismutase, malondialdehyde and catalase in the hepatic tissue were measured using specific
enzyme linked immunosorbent assays and spectrophotometry. High fat diet significantly reduced glutathione peroxidase
activity (P = 0.04) in the hepatic tissue. Vitamin D treatment led to a significant reduction in hepatic tumor necrosis factor
concentrations and increased superoxide dismutase activity in the group receiving normal diet + vitamin D (P = 0.001), but
vitamin D had no effects on glutathione peroxidase or catalase activity. Hepatic malondialdehyde concentration at baseline
or after vitamin D administration was not significantly different. Vitamin D improved hepatic oxidative stress and inflammatory
marker in high fat diet-induced obesity. |
