| Metformin is one of the first line drug of treatment in type 2 diabetes. Various analytical methods have been reported for determination of metformin in plasma. We determined metformin in human plasma sample using a dispersive liquid-liquid microextraction (DLLME) procedure in combined with HPLC. Several effective factors in DLLME-HPLC-UV methods such as type of extraction and disperser solvents and their volume, centrifugation rate and pH were optimized. Under the optimum conditions, the method was validated over the concentration range 0.2-2.9 μg/mL in human plasma. Inter-day and intra-day precisions were less than 10% where the acceptable levels are less than 20%. Recoveries of 94-104 were achieved. Furthermore, the validated method was successfully applied for the determination of metformin in spiked plasma samples. The combined DLLME with HPLC is a simple, low cost and fast determination method for analysis of
metformin in human plasma. |
