| خلاصه مقاله | Due to the negative impact of reactive species (including free radicals) on humans and animals, the investigations to find effective substances (antioxidants), which protect living organisms against their damaging influence are carried out throughout the world. Astaxanthin (ATX) is an antioxidant that shows efficient scavenging ability and represents an interesting candidate in the development of new therapies for preventing and treating oxidative stress-related pathologies. However, its high lipophilicity and low thermolability often limits its antioxidant efficacy in biomedical applications. Here, we developed a formulation of chitosan-g-poly(lactide)(CS-g-PLA ) micellar nanoparticles to protect astaxanthin’s antioxidant activity. Polylactide and chitosan (CS) are biodegradable and biocompatible materials that widely been used as drug delivery vehicles. To prepare CS-g- PLA, the solubility of CS in organic solvents was improved by the complexation of CS with sodium dodecyl sulphate (SDS) simply by mixing acidic solutions of CS and SDS. After purification, graft copolymerization of lactide (LA) on CS backbone was carried out using ring opening polymerization. The obtained solution was purified via precipitation with tris buffer and washing with distilled water respectively and finally it was freeze-dried. The chemical structures of the copolymers were confirmed by Fourier transform infrared spectroscopy (FT-IR), 1H nuclear magnetic resonance (1H NMR) and thermogravimetric analysis (TGA). The efficiency of the developed nanoparticles would be evaluated for the encapsulation of astaxanthin as a potent antioxidant agent. |
