خلاصه مقالهBackground and Objective: Morphine is the most widely used analgesic for management of pain in the patients suffering from cancer. Tolerance to its analgesic effects, is one of the challenges in chronic administration of this opioid. The aim of this study was to investigate the effect of silymarin on the tolerance to analgesic effects of morphine in mice with skin cancer. Materials and Methods: Forty two cancerous male Albino-Swiss mice were divided into six groups. Two noncancerous groups were also studied (one as the control group and the other received morphine). To induce skin tumor, a single dose of 7,12-Dimethylbenz[a]anthracene (DMBA) was applied topically. After 7 days, croton oil (1mg in 250μl acetone) was administered twice a week topically for 16 weeks. Pain latency time was assessed using hot-plate test and tumorigenesis was controlled daily for 30 weeks. The normal and cancerous mice were treated with morphine (20 mg/kg/day). The control groups received saline in the same manner. Three groups of mice received silymarin (50, 100, 150 mg/kg) in addition to morphine. The last group received polyethylene glycol as the solvent for silymarin. Results: We observed that silymarin (100, 150 mg/kg) postponed the tolerance 7 and 12 days respectively. Conclusion: Our data suggest that silymarin may be able to delay the morphine tolerance in skin-tumor bearing patients. Further clinical studies are required to prove this.
