| خلاصه مقاله | Introduction: Early stage cancer detection could reduce breast cancer death rates significantly and is imperative for effective staging and evaluate cancer treatment response. To achieve this goal, Molecular imaging (MI) is a potentiated technique in order to detect biological processes at the cellular and sub cellular levels by using targeted imaging agent that can bind targets of interest with high specification. Method: The surface modifications of gold nanoparticles (Au-NPs) were done with long PEG (HS-PEG-CH3O) and short PEG (HS-PEG-COOH) chains to enhance their stability and capacity for immobilization of anti CD24 antibodies. CD24-AuNPs PEGylated were characterized via different techniques and MTT assay was carried out to assess the biocompatibility of the NPs. Obtained contrast agent was implemented in the targeted CT imaging based on in vitro and in vivo studies of breast cancer. Result: The CD24-AuNPs appeared as spherical nanoparticles with 100 nm diameters also showed biocompatibility without toxicity in a studied concentration range. Study on CT x-ray attenuation property revealed that the attached CD24 to the cell surface of PEGylated Au-NPs could significantly enhance the cells CT number (47.68 HU in 4T1, while it was 16.61 HU in CT26). Further, the tumor region in model of xenograft tumor has higher density compare to the control groups, 60 min after injection (45 Hu vs.91 Hu). Discussion: the synthesized nanoparticles can be used as computed tomography contrast agent to identify the tumor cells with higher expression of CD24 at the early stages more efficiently in comparison to the other imaging methods. |
