| خلاصه مقاله | Calcium carbonate is an important inorganic material and typical bio-mineral, which is widely used by several industries such as construction, medical, paint, plastic,rubber, ceramic, glass, steel, oil refining and paper. Microorganism-based approach is a new producing method for Calcium carbonate which it happenese through urease activity of Sporosarcina pasteurii during the growth of the microorganism with the surrounding environmental media. The yield of urease activity plays an important role in final material production and it is affected mostly by Sporosarcina pasteurii growth conditions such as pH, temperature, cultivation medium.
Here, for optimization of growth condition, Sporosarsina pasteuri, with No. 1645 (DSM 33) is provided from the Persian type culture collection (PTCC). In order to optimize the growth condition to reach the maximum urease activity, the effect of three different factors of nitrogen source(Ammonium chloride, Ammonium nitrite, ammonium hydrogen phosphate), pH( 8,8.5 and 9) and temperature (25,30,35 oC) was investigated in three levels.
The results showed that the highest urease activity is in liquid cultivation containing Ammonium chloride as a nitrogen source ,pH of 8.5 and temperature of 30 oC. Moreover, the optimization of Ammonium chloride concentration demonstrated the urease activity is most preferable in 10 gr/L. |
