| خلاصه مقاله | Over the past few decades, there has been comprehensive progress in terms of improving a more effective chemotherapeutic regimen for targeted Breast cancer therapy. To overcome cancer resistance to monotherapies, drug combinations of two or more compounds with different mechanisms of action are developed. We explored 2 agents with promising preclinical data in in-vitro assays. Firstly, Erlotinib used as a tyrosine kinase inhibitor. This agent is a promising class of Anti-angiogenic agents which has attracted a lot of attention in combinational therapy. The second drug was metformin. It is generally a safe and effective medication in type 2 diabetes. Several studies represent its efficacy in cancer study. The thermal decomposition process was used for synthesizing the magnetic core. To obtain a mesoporous silica shell, NaOH, acetone solution of APTES, NH4NO3, and NaBH4 used. The reaction mixture was then stirred at room temperature overnight. The success of this synthesis was confirmed by DLS, TEM, SEM, FT-IR and VSM methods. In the next step, The Encapsulation efficiency (EE %) and Loading Capacity (LC %) of each drug was calculated. the pH-responsive release profile of Metformin and Erlotinib from MSMNPs in PBS (pH 5.4, 6.4 or 7.4) was plotted. The experiments were repeated three times. The results indicated that the release of drug molecules and the release rate was greatly dependent on the pH value. For Cytotoxicity assay firstly we determined ic50 of both-agent including metformin and erlotinib in 2D and 3D MCF-7 cell lines. To define synergistic combinations of these drugs we use Combenefit and CompuSyn software. All statistical analysis was performed using SPSS 17.0.0 software. Finally, we choose the combinations which include less molar concentration of Erlotinib that may have less adverse effects.
