| خلاصه مقاله | Introduction and Background: Cardiovascular diseases (CVDs) are one of the most important causes of morbidity and mortality and their risk factors are high blood pressure, coronary thrombosis, strokes and renal failure. Quantification of CVD drugs such as calcium channel blockers in biological samples is essential in therapeutic drug monitoring (TDM). Verapamil hydrochloride belongs to the calcium channel blockers (CCBs) used to treat a multitude of CVDs such as hypertension, supraventricular tachyarrhythmias, hypertrophic cardiomyopathy and angina pectoris. A combined microextraction and separation method is presented for the determination of verapamil in EBC which is a promising non-invasive biological component for monitoring drug concentrations. Methods: In this work, dispersive liquid–liquid microextraction (DLLME) procedure was employed for pre-concentration of verapamil in EBC samples and followed by determination with high-performance liquid chromatography- ultraviolet detection (HPLC -UV) at 200 nm. The method was developed and validated for quantitation of low level of verapamil in patient's EBC samples. Results: Under the optimal conditions, the method indicate good stability and selectivity. The proposed method was applied for analysis of verapamil in real samples successfully. Discussion and Conclusion: A reversed-phase HPLC-UV method was developed and validated for analysis of verapamil in human EBC samples. The suggested method can be employed as a routine analytical procedure for TDM and clinical studies. |
