| خلاصه مقاله | Oxaliplatin (OXP) is one of the effective apoptotic therapeutics to treat different types of solid tumors; however, it has a limited bioavailability and cellular uptake due to its low permeability. To tackle this, we aim to design different cell-penetrating peptides(CPPs) conjugated with cholesterol(COL) to surface modify the oxaliplatin liposomes. This novel strategy improves the intracellular uptake of OXP without causing any immunogenic response. CPPs were synthesized by the solid-phase peptide synthesis method. Peptide-COL conjugates were prepared via covalent binding of peptide and COL. CPPs and peptide-COL conjugates were characterized using Nuclear Magnetic Resonance (NMR). Modified OXP liposomes were prepared by the ethanol injection method and were characterized using zeta-sizer. Different CPP Modified liposomes uptake was evaluated in breast cancer cell lines (MCF-7) by flow cytometry. Cytotoxicity of CPP- modified OXP liposomes was evaluated by MTT assay. Amongst all sequences, R8-Liposome possessed the highest drug loading efficiency (36%), where the lowest drug loading efficiency (19%) belonged to W4R4-Liposomes. All modified liposomes had an acceptable size (<200 nm). The selected CPPs and peptide-MTX conjugates did not show any cytotoxicity or immunogenicity. The CPP- modified liposomes showed a suitable impact on uptake and apoptosis of breast cancer cell lines MCF-7. Thus, this novel drug delivery system is represented as promising carriers for improving the intracellular uptake of OXP liposomes with low systemic toxicity in breast cancer treatment. and cytotoxic effects on normal cells. |
