| خلاصه مقاله | Introduction and background: The genus Pseudomonas, belonging to the family Pseudomonadaceae, was first proposed by Walter Migula. (1894). They have essential roles in decomposition of organic matter, in the nitrogen cycle and improvement of plant growth and are also known as plant, animal and human pathogens. (1) During a survey of bacteria with biodegradation ability of polycyclic aromatic hydrocarbons (PAHs) in Caspian Sea, a bacterium was isolated. The isolated strain, TBZ2T was subjected to a polyphasic taxonomic analysis. This strain is considered to represent a novel species of genus Pseudomonas.
Methods: DNA was extracted from isolated strain TBZ2T. The 16S rDNA was targeted for amplification by polymerase chain reaction. The amplified fragment was purified and sequenced and results were analyzed using Chromas Pro software. Phylogenetic tree was constructed using neighbor-joining method in MEGA 6 software. Biochemical tests containing API 50CH, API 20E and API 20NE kits and some lab-made tests were performed.
Results: The isolated bacterium is a Gram-negative, rod shaped, non-motile Which produce cream colored colonies on marine agar medium with circular shape, convex angle and entire edges. The cells grow at 0-8.5% w/v NaCl with optimum growth in 2-5% NaCl and are able to grow at 10-45 ºC, pH range of 3-10 with the efficient growth at 30 °C and pH of 5. Analysis of the 16S rRNA gene sequences revealed that strain TBZ2T belonged to the genus Pseudomonas. Similarities between strain TBZ2T and closely related type strain, obtained by using the EzTaxon database were 98.79% with Pseudomonas. oleovorans subsp. oleovorans DSM 1045(T).
Discussion and Conclusion: The results of whole findings including 16S rRNA gene sequence, morphophysiological and biological tests justify a distinction for strain TBZ2T on species level in Pseudomonas genus. Therefore, strain TBZ2T is suggested to be a novel species which named Pseudomonas Khazarica TBZ2T. |
