| خلاصه مقاله | Introduction and background: Polycyclic Aromatic Hydrocabons (PAHs), are kind of genotoxic contaminations with high concentrations in many industrial sites and are considered as one of the environmental concerns since stability against degradation and potential of bioaccumulation in food chain. Bacteria are specially involved in their biodegradation among microorganisms which are being progressively isolated from marine habitats. This study was aimed to compare biodegration potential of five bacterial isolates from Caspian Sea qualitatively and quantitatively.
Methods: For qualitative tests for each isolate, PAH-supplemented ONR7a solid media was used. Three stock solutions were made by dissolving Naphthalene, Phenanthrene and Anthracene repectively in Acetone with concentration of 1 mg/ml and were added in three ONR7a media before solidification, in order to make supplemented ONR7a solid media with concentration of 200 ppm for Naphthalene, Anthracene and Phenanthrene for each of them. Positive control was performed by glucose supplemented ONR7a medium with concentration of 200 ppm. Three ONR7a solid media was made as test, positive control and negative control plates for each of PAH to be qualitatively tested for being biodegraded. To evaluate the Naphthalene, Phenanthrene and Anthracene degradation potential of bacterial isolates, these substrates were provided with a concentration of 50 ppm as the sole source of carbon. Then, the samples were incubated. Screening was done through spectroscopic absorption of the samples at λ max of each PAH and through calculating residual PAH concentration in each day of incubation by means of calibration curve drawn based on standard samples.
Results: Qualitative Biodegradation Tests represented that, stains SFR2 (=SS1) belonging to the genus Photobacterium and SS2 belonging to the genus Pseudomonas can degrade Naphthalene, Phenanthrene and Anthracene more efficient than others. So these strains were subjected for Quantitive Biodegradation Tests. Bacterial growth curve for two isolates and their potential to reduce PAH concentration in medium was obtained by quantitative measurement.Discussion and Conclusion: By taking into consideration all results, it can be concluded that bacteria of Caspian Lake have an efficient role in biodegradation of PAHs. |
