| خلاصه مقاله | Introduction and background: One of the major methods to reduce various pollutants is applying biologic method which is known as bioremediation. Anthracene is a tricyclic aromatic hydrocarbon that is found in high concentrations in polycyclic aromatic hydrocarbon (PAH)-contaminated sites. Bacteria are specially involved in PAH biodegradation among microorganisms which are being progressively isolated from marine habitats. So, this study was aimed to analyze the existence of PAH-degrading bacteria phenotypically and genotypically in Caspian Sea. Microbial selection was based on ability to use Anthracene as sole organic carbon source. Methods: Two samples were taken from estuary of Siahrood river and Fereydunkenar beach. Isolation of aquatic bacteria were performed on marine agar and sea water medium. In order to enrich Anthracene-degrading bacteria, Anthracene supplemented- ONR7a medium was used which dosen't contain any organic carbon source itself. Anthracene-degrading isolates were subjected for 16S rRNA sequencing. Phylogenetic tree was constructed using neighbor-joining method in MEGA6 software. Biochemical tests containing API 20E and API 20NE kits and some lab-made tests were performed Results: Twenty bacterial isolates were isolated from these water samples on marine agar and sea water medim. six bacterial isolates were able to grow on supplemented medium described above: SF2, SFR1, SFR2 (SS1), SFR3 and SS2. The 16S rRNA gene sequence of each isolate was analyzed based on NCBI and EZtaxon data bank in order to find out their phylogenetic position and the respective phylogenetic tree was constructed. Based on phylogenetic results, it is recommended that the isolates belong to Shewanella, Photobacterium, Mycobacterium and Pseudomonas. Discussion and Conclusion: By taking into consideration all results and searching references, it can be concluded that bacteria of Caspian Lake have an efficient role in biodegradation of PAHs. By separating the PAH degrading bacteria from Caspian Lake, it has been clarified that part of the purification process of the lake is done by these bacteria. |
