| خلاصه مقاله | Effect of Nigella sativa and thymoquinone on endothelin level in ovalbumin-sensitized guinea pigs Introduction: Nigella sativa extract has been used for treating asthma in traditional Medicine, but its exact mechanism is not obvious. Previous studies showed that thymoquinone, the main constituent of Nigella sativa, has bronchodilatory effects. So this study was proposed for determining the effect of hydro-ethanolic extract and thymoquinone on endothelin 1 level in bronchoalveolar lavage (BAL) and pulmonary tissue of experimental asthmatic guinea pigs. Method: 60 guinea pigs of both sexes were randomly divided to control (group C), animal model of asthma (group S), sensitized groups drinking water containing 0.125 g/dl Nigella sativa extract (group S+LNS), and 0. 25g/dl Nigella sativa extract (groupS+HNS) and sensitized groups drinking water containing 20μMthymoquinone (group S+LTQ) and 40μMthymoquinone (group S+HTQ). Results: The mean value of endothelin in BAL and lung tissue of sensitized group was significantly higher than in group C (p<0.001 to p<0.05).The mean value of endothelin in BAL and lung tissue in all pre-treated groups were significantly lower than S group (p<0.05).There was not any significant difference between groups S+LNS and groupS+HNS and between group S+LTQ and group S+HTQ.The mean value of endothelin in BAL and lung tissue in group S+LNS were significant in comparison with S+LTQ(p<0/01) but there was no significant difference between S+HNS and S+HTQ groups. Conclusion: It confirmed Nigella sativa and thymoquine have a reducing effect on endothelin level in both BAL and lung tissue in sensitized guinea pigs. |
