| نوآوری و ضرورت انجام تحقیق | Several methods have been used for the determination of vancomycin in bulk, dosage forms and plasma samples. The important characteristics of an analytical method to be used in clinical and diagnostic are its reliability, low cost and rapid determination of analyte concentration in small volumes of biological fluids. Unfortunately, the most of existing methods are expensive, complicated analysis systems, time consuming, cumbersome and hard operation. The aim of this study is to develop an easy and low-cost method for determination of vancomycin in biological samples such that has the potential to improve outcomes at the point of care. |

| چکیده انگلیسی طرح | Vancomycin is recommended intravenously as a treatment for complicated skin infections, bloodstream infections, endocarditis, bone and joint infections, and meningitis caused by methicillin-resistant Staphylococcus aureus. Vancomycin is also recommended by mouth as a treatment for severe Clostridium difficile colitis. Level monitoring of vancomycin in body fluids is necessary due to the drug's biexponential distribution, intermediate hydrophilicity, and potential for ototoxicity and nephrotoxicity, especially in populations with poor renal function and/or increased propensity to bacterial infection. By considering this fact, the monitoring of vancomycin concentration in the biological samples of patients will be important. The main aim of this method is development of an optical method for vancomycin analysis. The proposed method is based on quantum dots. A fluorescence instrument is applied for drug monitoring with proposed method. Several parameters affecting the analytical signal including: quantum dots type, reagent concentration, reaction time etc. will be studied and optimized. Response surface methodology was employed to study the influence of reaction parameters affecting the analysis by considering the interactive effects between independent variables. After optimization and validation of the proposed method, the calibration graph in the biological matrices will be made. Then, the validated method will be used for determination of vancomycin in the biological samples of patients' receiving drug. |
