A laser based technique for the solubility study of ketoconazole in the solvent mixtures at various temperatures

خلاصه روش اجرا: Modeling of drug solubility and its experimental determination using automated techniques have great importance in the pharmaceutical industry. Ketoconazole solubility was measured in a solvent mixtures at various temperatures using an updated laser-based monitoring combined with gravimetric measurement. Unlike a shake-flask method, in this system, the under-saturated solvent mixture reaches the saturation point by gradual ketoconazole powder addition using a robotic arm. The solubility value is obtained from the ketoconazole powder mass initially placed in the inject cell, and the mass of the residual ketoconazole in the inject cell after equilibration.

| نوآوری و ضرورت انجام تحقیق | The poor aqueous solubility of a number of azole antifungals (e.g. ketoconazole) is very well known. So, in this work, we will measure its solubility at different cosolvency mixtures. This will help us expand the current knowledge about the effect of cosolvents type on thermodynamic aspects of drug dissolution including the roles of crystal lattice structure (which is a solute property alone and remains the same regardless of the solvent type) and the solute-solvent interaction forces (which varies with the addition of cosolvents). The expanded solubility database, will have a big impact in identifying the relative importance of these two solubility factors for different molecular structures and/or structural fragments. In addition, various temperatures will also be investigated which will aid to characterize the effect of temperature in cosolvency problems. |
