| نوآوری و ضرورت انجام تحقیق | According to current literature, the earliest pathophysiology involved is the aggregation of amyloid beta forming the amyloid plaques within the brain. Nevertheless, to date, there is no report for the investigation of Aβ in respect to its protein network entropy. In our previous works – yet unpublished – we have proposed a mathematical theorem based on the information theory. Also, we have devised a novel methodology based on Shannon entropy and coined a new unit terminology for the calculation of entropy in proteins, i.e. “pit”. By the means of network analysis, in this study, we aim to evaluate the protein network of amyloid beta aggregation in accordance with our novel methodology. |

| چکیده انگلیسی طرح | A review of the literature will be done to understand and indicate the exact proteins involved in the pathophysiology of amyloid beta aggregation. Required information for all included proteins will be extracted from UniProt database. Then, their number of interactions will be specified via a validated comparison between IntAct, BioGrid, DIP, MINT, HPRD databases. This process will be facilitated using PICKLE meta-database which is available online. Then, for all included proteins, and interacting proteins, Shannon entropy will be calculated in pits unit, according to our methodology. Then, hub proteins, and non-hub proteins will be specified by the use of ANOVA post hoc statistical analysis, and the type of network will be identified for future research on network analyses. |

Shannon entropy: The mathematical modeling of uncertainty in a system, and the measured randomness thereupon which was proposed by Claude Shannon in 1948 (11). Protein network: A set of orchestrated proteins whose funtions result in a biochemichal outcome is called protein network. These proteins interact with one another in the network (14). Amyloid beta: Peptides with 36-43 amino acids that are considered to have a key role in the pathogenesis of Alzheimer’s disease (8). |
| ذینفعان نتایج طرح | 1. Neuroscientists, neurologists and psychiatrists: They will all have a much better understanding of the pathogenesis behind the AD. 2. AD patients: They are definite stakeholders of current study, as it may lead to next studies on drug targeting based on entropy-based network analysis. 3. Health system: The results of this study may reduce the substantial burden of AD disease promoting public health. |
