| خلاصه مقاله | Abstract Introduction: Protein acylation is a posttranslational modification in which an amino acid residue of a protein is acylated by a fatty acid. In spite of extremely complicated molecular methods, global protein acylation can be profiled following the hydrolysis of fatty acyl groups from cellular proteins. Aim: The aim of this study was to develop a method for the analysis of global protein acylation based on gas-liquid chromatography (GLC) technique. Methods: Total protein was extracted from human hepatocellular carcinoma (HepG3) cell line. Protein sedimentation and extensive wash were combined with differential acyl hydrolysis using sodium hydroxide (NaOH), hydroxylamine (NH3OH) or hydrochloric acid (HCL). GLC was used to analyze the fatty acid composition changes of released lipids. Along with the expression assay of stemness markers, the suggested method was applied for the determination of the effect of chemical inhibition of monounsaturated fatty acid (MUFA) synthesis on global protein acylation. Results: Only NaOH could release significant amounts of stearate. The highest release of palmitoleate was seen with NH3OH. The amount of myristate was significantly higher than other fatty acids released by HCL. Treatment with a chemical inhibitor of monounsaturated fatty acid synthesis led to decreases in palmitoleate and oleate acylation of HepG3 cellular proteins, which were accompanied with significant reductions in the gene expression of Oct3 (-37%, p<7176) and Sox3 (-37%, p=7173). Conclusion: GLC based analysis of global protein acylation affords a semi-quantitative method assessing the changes in the protein acylation profile during stem cell differentiation and reprogramming. |
