خلاصه مقالهIntroduction Cytokines play fundamental roles in the pathogenesis of BD. Interleukin 27 (IL-27) is a pro-/anti-inflammatory cytokine which acts as a mediator between the innate and adaptive immune systems. IL-27 recently introduced in the pathogenesis of BD. The purpose of this study was to investigate a possible association between rs153109 and rs181206 polymorphisms of IL-27 gene and susceptibility to BD. Materials and methods The study group consisted of 50 Iranian patients with BD, 29 (58%) men and 21 (42%) women and one hundred healthy individuals. All patients fulfilled the International criteria of BD (ICBD). The study protocol was approved by the Ethic Committee of Tabriz University of Medical Sciences and written informed consent was received from participants. Genomic DNA was extracted from the whole blood collected in tubes containing EDTA using the standard salting-out method. The rs153109 polymorphism in IL-27 gene was analyzed by the Polymerase Chain Reaction–Restriction Fragment Length Polymorphism (PCR–RFLP) method, utilizing the specific primers. The IL-27 rs181206 polymorphism was genotyped using Amplification Refractory Mutation System-Polymerase Chain Reaction (ARMS-PCR) method. Results Comparison of the genotype frequencies of IL-27 rs153109 (A/G) polymorphism revealed statistically significant differences between BD patients and normal groups. Similarly, allelic frequencies also exhibited significant changes in the case-control distribution. Our results showed significantly higher frequency of AA genotype (40% versus 22%; p = 0.034) and lower frequencies of GG and AG genotypes (respectively: 12% versus 25% and 48% versus 53%) in BD patients as compared to the healthy individuals. No significant differences were observed in allele and genotype frequencies of the rs181206 SNP between BD patients and controls.
